Harmonix infused a lot of soul into that game. I'll be surprised if John Tam can manage to keep that up when working with Neversoft, unless Neversoft is also comprised mostly of musicians.
Guitar Hero will go the way of DDR - lots of releases to get new music, attempts at innovation that go horribly wrong (Hands & Feet mode, quest mode, Ray Charles tracks in a dance game).
Not that Harmonix is immune from that. They're up to eight versions of Karaoke Revolution, with some good innovations (duets) and some bad (integration of dance pads). Then there's the new trend towards cross-licensing (American Idol, Country Music Television) which is only going to get worse now that they're owned by Mtv.
