It's not about aiming or twitch reflexes. Those are pretty much taken out of the game entirely, because the actual kill(offence) or stun(defense) mechanics are prompted by the game (with an icon over the targets head). So with that out of the equation, it's all a mind-game. It's about hiding without looking guilty, chasing and approaching a target without letting them know you're even there.
Every kill is like a long, suspenseful cold war, before the first shot is fired. From there, it's classic AC. Either an amazingly cool kill sequence, or an intense chase through the streets or over the roof-tops.
I love the Team modes, because you can combine abilities in really cool ways, and rack up the bonuses for impressive kills. Casually walking past a target and hitting the poison button, and then walking away as they drop dead a few seconds later is soooooooo satisfying. So is tossing a throwing knife at a runner right before he gets to a chase-breaker, and then shanking him. :D

I've been in matches where a guy has scored something like 13 assassinations, or something ludicrous like that, but I'm still ahead of him on the leaderboard because while he was running around, getting noticed, and just shanking guys in the open, I was keeping silent, using abilities, and teaming up for kills with teammates for HUGE kill bonuses.
13 hundred point kills or 4 800 point kills? However you wanna play it man. :D
