It is a charity event raising money for Child's Play! The more money that is donated, the more levels of various Mario platformer games they will complete, all streamed live over the internet. Last year they played for more than 3 days straight and raised $29,082!
A fan put together a little intro video for people who haven't seen the past Mario Marathons, as an introduction to the event and the people behind it.
